TransIT®-Jurkat Transfection Reagent: Optimised Delivery for T-Cell Lineages
The TransIT®-Jurkat Transfection Reagent is a specialised formulation designed to overcome the inherent challenges of transfecting Jurkat cells and associated lineages. While Jurkat cells (human T-lymphocytes) are a critical tool in immunological research, they are notoriously difficult to transfect with standard reagents. This reagent is specifically optimised to deliver exceptional transfection efficiency for plasmid DNA in these hard-to-transfect cells, as well as other hematopoietic lines such as THP-1 and RAW264.7.
The reagent employs a high-efficiency technology that maintains low cellular toxicity, ensuring that experimental results are not compromised by cell death or stress. It offers a streamlined workflow that is fully compatible with serum-containing media, meaning no medium changes are required post-transfection. Whether performing transient gene expression studies or generating stable cell lines, TransIT-Jurkat provides the reproducibility and simplicity expected from the trusted TransIT series.
Key Benefits:
- Lineage Specificity: Optimised for Jurkat. Specifically developed to achieve high efficiency in Jurkat cells (acute T-cell leukaemia) and other difficult hematopoietic lines like THP-1 and RAW264.7.
- Experimental Integrity: Low Toxicity. The gentle formulation minimises cytotoxicity, preserving cell health and morphology for reliable data.
- Workflow Efficiency: Serum Compatible. Designed for use in complete growth medium; eliminates the need for medium changes to remove transfection complexes.
- Versatile Use: Transient & Stable. Supports both short-term transient expression assays and the generation of stable cell lines.
- Consistent Performance: Reproducible Results. Delivers consistent transfection efficiency, allowing for dependable comparison between experiments.
Technical Insights
Protocol Optimisation
Transfection efficiency can be fine-tuned by adjusting the reagent-to-DNA ratio. The recommended starting point is 3 µl of TransIT-Jurkat Reagent per 1 µg of DNA. Researchers can optimise performance for specific Jurkat subtypes by titrating the reagent from 1–5 µl per 1 µg of DNA. In some cases, doubling both the amount of reagent and plasmid DNA can further increase protein expression.
