RNase-Free DNase I Kit: Complete Removal of Genomic DNA for Sensitive RNA Applications
Norgen’s RNase-Free DNase I Kit is an essential accessory for researchers requiring RNA of the highest purity. While Norgen’s spin-column RNA isolation kits are known for their exceptional ability to exclude genomic DNA, certain ultra-sensitive applications—such as low-copy qPCR, RNA-Seq, and array analysis—demand the absolute removal of all trace DNA contamination. This kit provides a guaranteed RNase-free source of DNase I, ensuring the integrity of your delicate RNA samples while effectively digesting residual genomic DNA.
The kit is highly versatile, offering optimized protocols for two distinct workflows: On-Column Digestion, which integrates seamlessly into the wash steps of Norgen’s RNA purification kits for a rapid, “zero-step” removal; and In-Solution Digestion, for treating RNA samples that have already been eluted. Both methods utilise the provided Enzyme Incubation Buffer to maximise enzyme activity without compromising RNA yield.
Key Benefits:
- Guaranteed RNase-Free: Protect your samples. The enzyme and buffers are rigorously tested to be free of ribonuclease activity, ensuring that your RNA remains intact and undegraded during the digestion process.
- Seamless On-Column Protocol: Save time. Perform the digestion directly on the spin column during the RNA binding/washing steps. This efficient workflow removes DNA before elution, eliminating the need for post-extraction cleanup steps.
- Versatile Application: Flexible use. Includes a robust protocol for in-solution digestion, allowing you to “rescue” previously isolated RNA samples that show evidence of DNA contamination.
- Optimised Buffer System: High efficiency. The included Enzyme Incubation Buffer is specifically formulated to support maximum DNase I activity in solid-phase (column) applications, where standard buffers often fail or reduce RNA binding efficiency.
Protocol Versatility: On-Column vs. In-Solution
The kit is designed to adapt to your specific workflow needs:
- On-Column Digestion: Applied after binding RNA to the column but before the final wash. The DNase I solution gently digests DNA bound to the resin, and the enzyme is subsequently washed away, leaving pure RNA for elution.
- In-Solution Digestion: Used for total RNA that has already been eluted. Following digestion, the RNA can be cleaned up using Norgen’s specialised kits to remove the enzyme and buffer salts. We recommend:
- RNA Clean-Up and Concentration Micro-Elute Kit (for elution in as low as 8 µL)
- RNA Clean-Up and Concentration Kit (Standard Format)
- RNA Clean-Up and Concentration Kit (96-Well) (High Throughput)
