RNA/DNA Purification Kit – 48700: Sequential Total RNA and Genomic DNA Extraction
The RNA/DNA Purification Kit provides a rapid, streamlined method for sequentially isolating and purifying total RNA and genomic DNA from a single sample. When working with precious, difficult-to-obtain, or small samples such as biopsy materials or single cell foci, splitting lysates can lead to inconsistent results and variability. This kit overcomes these challenges by allowing you to extract both macromolecules without fractionating the initial source material, ensuring that your genome and transcriptome analyses are perfectly correlated.
Utilising advanced spin column chromatography, the purification process relies on a proprietary resin acting as a highly efficient ion exchanger. The procedure begins by lysing the cells or tissue with the provided Buffer SKP. This single lysate is first passed over a DNA column to selectively capture genomic DNA. Ethanol is then added to the flowthrough, which is subsequently loaded onto an RNA spin column to bind total RNA, including microRNAs, while proteins are washed away. In under 30 minutes, both DNA and high-purity RNA—ranging from large mRNA down to microRNAs (<200 nt)—are eluted without the need for hazardous phenol extractions or precipitation steps.
Key Benefits:
- Single-Sample Efficiency: Eliminate lysate splitting. By sequentially isolating RNA and DNA from the exact same sample, you reduce technical variability and guarantee reliable downstream analysis.
- Comprehensive RNA Recovery: Purify all sizes of RNA. The specialised resin ensures the capture of total RNA, from large mRNA and ribosomal RNA down to microRNA (miRNA) and siRNA, without bias.
- Rapid Protocol: Complete extractions in 30 minutes. The robust two-column system bypasses time-consuming phenol extractions and precipitation methods to deliver highly streamlined laboratory workflows.
- Broad Sample Compatibility: Optimised for diverse inputs. Extract nucleic acids effectively from cultured animal cells, fibrous tissues, blood, bacteria, yeast, fungi, and plant tissues.
Technical Insights
The core technology behind the RNA/DNA Purification Kit lies in its unique dual-column fractionation. By isolating genomic DNA first, the system prevents genomic contamination in the RNA fraction, ensuring the highest integrity for sensitive downstream applications such as microRNA profiling, mRNA knockdowns, and biomarker discovery. The lack of phenol or chloroform steps further guarantees that the extracted nucleic acids are strictly high-purity and immediately compatible with RT-qPCR, next-generation sequencing, and microarray analyses.
