Optimisation Guidelines
To ensure optimal performance, the TransIT-Oligo protocol provides specific starting conditions based on the type of oligonucleotide used. For 2’OMe RNA transfection, a starting concentration of 2 µM (final concentration) is recommended, with an optimisation range of 0.5–5 µM. For sDNA oligos, a lower starting concentration of 100 nM is suggested, typically ranging between 50–200 nM. The reagent volume can also be titrated (e.g., 2–5 µl per well in a 24-well plate) to balance efficiency with cell density.
