TTE-Glycerol Tolerant Buffer (20 X): Eliminate Band Distortion
National Diagnostics TTE-Glycerol Tolerant Buffer (20 X) is a specialised electrophoresis reagent developed to solve a specific challenge in DNA sequencing: the distortion of bands caused by the presence of glycerol. When samples containing high concentrations of glycerol—common in many enzyme-based sequencing reactions—are run using standard TBE, the result is often “smiling” or blurred bands. TTE-Glycerol Tolerant Buffer eliminates these artefacts, providing crisp, horizontal band migration.
This buffer utilises a Taurine-based chemistry to replace the Borate found in standard TBE. The interaction between glycerol and borate typically creates a negatively charged complex that alters the local conductivity and mobility of the DNA. By substituting Borate with Taurine, the TTE system remains inert to glycerol, ensuring uniform migration across the entire gel and significantly improving the accuracy of automated and manual sequence reading.
