pOET1 Transfer Plasmid: Efficient Baculovirus Vector

The pOET1 Transfer Plasmid is a highly efficient baculovirus transfer vector designed to facilitate the high-level expression of foreign genes in insect cells. Controlled by the powerful AcMNPV polyhedrin (polh) promoter, this plasmid drives robust protein production late in the infection cycle. Its streamlined design is significantly smaller (~4.6 kb) than many traditional transfer vectors, making cloning steps faster and less prone to errors.

Ideally suited for use with the flashBAC™ Baculovirus Expression System, pOET1 contains the essential AcMNPV and ORF1629 flanking sequences required for homologous recombination with the viral genome. This ensures the precise insertion of the expression cassette into the polyhedrin locus. The vector replaces the native polyhedrin coding sequences with a Multiple Cloning Site (MCS), allowing for the directional insertion of your gene of interest.

The pOET1 plasmid is engineered for performance and ease of use. It includes the Col E1 origin of replication (467–1056 bp) and Ampicillin resistance gene (1211–2071 bp) for propagation. The recombination sequences AcMNPV (2656–3316 bp) and ORF1629 (4035–4574 bp) flank the expression cassette. The Multiple Cloning Site (3317–3403 bp) is positioned immediately downstream of the Polyhedrin promoter (3404–3535 bp).
