TransIT-VirusGEN® Transfection Reagent: Scalable AAV & Lentivirus Production
TransIT-VirusGEN® Transfection Reagent is a breakthrough technology designed to revolutionise the production of recombinant Adeno-Associated Virus (AAV) and Lentivirus. Unlike reagents limited to specific formats, TransIT-VirusGEN is engineered for broad scalability, delivering consistent high-titer performance in both adherent and suspension HEK 293 cell platforms. This versatility makes it the ideal solution for workflows ranging from small-scale process development to large-scale biomanufacturing.
The reagent features a novel, animal-origin free formulation that enhances the delivery of packaging and transfer vectors (DNA) to the nucleus. By removing the need for media changes and complex wash steps, TransIT-VirusGEN streamlines the viral production process. It is fully compatible with chemically defined media and standard manufacturing protocols, offering a robust and reliable path to clinical-grade viral vectors.
Key Benefits:
- Dual Utility: AAV & Lentivirus. a single, powerful reagent optimised for the high-titer production of both AAV and Lentivirus vectors.
- Scalable Performance: Adherent & Suspension. Delivers consistent results across different culture platforms, from tissue culture dishes to bioreactors.
- Regulatory Friendly: Animal-Origin Free. Fully synthetic and animal-origin free, supporting seamless transition from R&D to clinical trials.
- High Efficiency: Reliable Titers. Enhances DNA delivery to maximise the assembly and release of functional viral particles.
- Streamlined Workflow: Chemically Defined. Compatible with various chemically defined and serum-free media formulations; no media changes required.
Technical Insights
Protocol Optimisation
For AAV generation, the recommended starting ratio is 2 µl of TransIT-VirusGEN per 1 µg of DNA, with an optimisation range of 1.5–4 µl. For Lentivirus production, similar ratios are employed to balance capsid production with cell viability. The reagent is designed to work effectively with various packaging mixes and transfer plasmids, allowing for flexible adaptation to existing manufacturing protocols.
