Protocol Optimisation
To maximise viral titers, the TransIT-Lenti protocol uses a specific reagent-to-DNA ratio. The recommended starting condition is 3 µl of TransIT-Lenti Reagent per 1 µg of total DNA (transfer vector + packaging mix). Complexes are formed in serum-free medium (e.g., Opti-MEM®) and incubated for 10 minutes before being added to cells. Viral supernatants are typically harvested 48 hours post-transfection.
