Protocol Optimisation
To maximise knockdown efficiency, the TransIT-siQUEST protocol allows for optimisation based on the specific cell type. For a standard 24-well plate, the recommended starting condition is 1.5 µl of reagent per well. However, researchers can fine-tune performance by titrating the reagent volume between 0.5 µl and 3 µl. The recommended starting siRNA concentration is 25 nM, with a typical working range of 10–50 nM.
