Protocol Optimisation
To achieve optimal knockdown efficiency, the TransIT-TKO protocol offers flexible optimisation guidelines. For a standard 24-well plate, the recommended starting condition is 2.5 µl of reagent per well with a final siRNA concentration of 25 nM. Researchers can further optimise delivery by testing reagent volumes between 1–4 µl. When performing co-transfection, it is recommended to synchronise the formation of DNA and siRNA complexes before adding them to the cells.
