Description

Label IT® Plasmid Delivery Control, Cy®3: Track and Optimise DNA Delivery

The Label IT® Plasmid Delivery Control (Cy®3) enables researchers to directly visualise and optimise plasmid DNA delivery in a wide range of experimental setups. This control features a 2.7 kb circular plasmid covalently labeled with the bright Cy3 fluorophore, serving as a reliable marker for tracking DNA uptake and localization within cells. It is an invaluable tool for troubleshooting transfection protocols and confirming successful delivery prior to functional analysis.

Designed for versatility, this control is suitable for both large-scale in vitro transfections and in vivo applications. Its ready-to-use formulation allows for simple integration into existing protocols, whether using chemical transfection reagents or electroporation. In vivo, it facilitates the monitoring of plasmid distribution following systemic or local administration, such as hydrodynamic tail vein injection.

Key Benefits:

  • Visual Tracking: Cy3 Labeled. Enables bright fluorescent monitoring of plasmid DNA delivery (Excitation: 550 nm, Emission: 570 nm).
  • Functional Mimic: 2.7 kb Plasmid. Mimics the size and topology of standard expression plasmids for relevant delivery tracking.
  • Versatile Use: In Vitro & In Vivo. Suitable for assessing transfection efficiency in culture or biodistribution in animal models.
  • Ready to Use: Convenient Format. Supplied at 0.5 mg/ml, allowing for direct addition to experimental workflows.
  • Broad Compatibility: Universal Application. Works effectively with standard broad-spectrum transfection reagents and electroporation systems.

Optimisation Guidelines

The intensity of the fluorescent signal may depend on cell type and transfection conditions. It is advisable to titrate the amount of control DNA to optimize visualization. Assessment of the signal is generally performed between 4 and 48 hours post-transfection. This 100 µg pack size is sufficient for transfecting approximately 50 wells of a 6-well plate or up to 10 mice.
