Description

Label IT® Plasmid Delivery Control, Fluorescein: Track and Optimise DNA Delivery

The Label IT® Plasmid Delivery Control (Fluorescein) is designed to assist researchers in visualising and optimising plasmid DNA delivery for both in vitro and in vivo applications. This control consists of a 2.7 kb circular plasmid covalently labeled with fluorescein, allowing for direct fluorescent tracking of DNA uptake and intracellular localization. By using this control, researchers can quickly assess transfection efficiency and optimise their protocols without the need for reporter gene expression assays.

Formatted for large-scale applications, this control is supplied at a ready-to-use concentration suitable for standard transfection or in vivo studies. It is fully compatible with broad-spectrum transfection reagents and electroporation, making it a versatile tool for evaluating delivery success in diverse cell types. In vivo, it can be used to monitor biodistribution following hydrodynamic tail vein injection or other delivery methods.

Key Benefits:

  • Visual Tracking: Fluorescein Labeled. Allows direct visualization of plasmid DNA delivery in cells and tissues (Excitation: 492 nm, Emission: 518 nm).
  • Functional Mimic: 2.7 kb Plasmid. Represents standard plasmid DNA in size and topology, ensuring relevant delivery characteristics.
  • Versatile Use: In Vitro & In Vivo. Optimised for assessing delivery efficiency in cell culture or tracking distribution in animal models.
  • Ready to Use: Convenient Format. Supplied at 0.5 mg/ml, ready for direct addition to transfection complexes or injection solutions.
  • Broad Compatibility: Universal Application. Compatible with standard transfection reagents and electroporation protocols.

Technical Insights

Optimisation Guidelines

Signal strength can vary based on transfection efficiency and cell type. For in vitro applications, it is recommended to titrate the amount of control DNA to determine the optimal level for visualization. Signal distribution is typically assessed between 4 and 48 hours post-transfection. For large-scale in vitro use, the 100 µg quantity provides sufficient material for approximately 50 wells of a 6-well plate (using 2 µg per well).
