TransIT®-mRNA Transfection Kit: High-Efficiency RNA Delivery
The TransIT®-mRNA Transfection Kit provides a highly efficient method for delivering messenger RNA (mRNA) and large viral RNAs into a broad range of cell types. By delivering RNA directly to the cytoplasm, this system bypasses the need for nuclear entry, resulting in faster protein expression and avoiding the risk of genomic integration associated with DNA transfection. This mechanism makes it an excellent choice for applications requiring rapid expression, such as vaccine development, CRISPR/Cas9 guide RNA delivery, and viral replication studies.
The kit is comprised of two components: the TransIT®-mRNA Reagent and the mRNA Boost Reagent, which work synergistically to maximise delivery and expression. The formulation is exceptionally gentle, maintaining high cell viability even in sensitive cells. Transfections can be performed in the presence of serum without the need for medium changes, ensuring optimal physiological conditions and simplifying the experimental workflow.
Key Benefits:
- Rapid Expression: Cytoplasmic Delivery. Delivers RNA directly to the cytoplasm for immediate translation, bypassing the nuclear barrier.
- High Efficiency: Boosted Performance. Includes a specialised mRNA Boost Reagent to enhance RNA uptake and expression levels.
- Superior Viability: Low Toxicity. Gentle formulation preserves cell health, essential for accurate functional assays and repeated transfections.
- Versatile Application: Diverse RNAs. Effective for delivering mRNAs, viral RNAs (2–10 kb), and CRISPR guide RNAs.
- Streamlined Workflow: Serum Compatible. Compatible with serum-containing media; eliminates the need for wash steps or medium replacement.

Protocol Optimisation
To achieve optimal results, the ratio of reagent to RNA can be fine-tuned. The standard protocol utilises both the TransIT-mRNA Reagent and the mRNA Boost Reagent. Researchers are encouraged to titrate the reagent volumes to determine the best balance for their specific cell type and RNA payload. The system is designed to work effectively with various RNA sizes and modifications.
