Protocol Optimisation
To achieve the best results, the reaction conditions can be optimised for specific keratinocyte subtypes. While the recommended starting condition is 3 µl of TransIT-Keratinocyte Reagent per 1 µg of DNA, researchers can titrate the reagent volume from 2–8 µl per 1 µg of DNA to determine the optimal ratio. For maximum efficiency, it is recommended to divide cells 18–24 hours before transfection to ensure they are actively dividing and reach approximately ≥80% confluence at the time of transfection.
