The buffer operates on the classic discontinuous buffer system principles. It provides the essential chloride and glycine ions required to stack and resolve proteins effectively. The inclusion of high-quality Sodium Dodecyl Sulphate (SDS) ensures uniform negative charge distribution across protein samples, maintaining their denatured state and facilitating migration based solely on molecular weight. Buffer Chemistry and Migration
Upon dilution to a 1X working concentration, this buffer typically yields a pH of approximately 8.3. This pH is critical as it regulates the ionisation state of glycine. In the stacking gel, glycine exists as a zwitterion (net neutral charge), moving slowly and creating a voltage steep that compacts proteins into a tight disc. Upon entering the resolving gel (higher pH), glycine becomes negatively charged and moves ahead of the proteins, allowing separation by size in the sieving matrix.
