TransIT-X2® Dynamic Delivery System: Versatile DNA & siRNA Delivery
The TransIT-X2® Dynamic Delivery System is an advanced, non-liposomal polymeric system engineered to enable high-efficiency transfection across a broad range of cell types, including difficult-to-transfect primary cells. Unlike traditional delivery methods restricted to specific macromolecules, TransIT-X2 is a multi-functional reagent designed for the delivery of plasmid DNA, siRNA, or the simultaneous co-transfection of both. This versatility makes it an essential tool for diverse applications, from gene expression and shRNA studies to promoter analysis and gene knockdown.
The system utilises a novel polymeric chemistry that combines high performance with ease of use. It allows for transfection in serum-containing medium without the need for post-transfection medium changes, significantly streamlining the experimental workflow. By eliminating the requirement for media replacement, TransIT-X2 reduces physical stress on cells and minimises the risk of contamination, ensuring that high efficiency is paired with optimal cell health and reproducibility.
Key Benefits:
- Multi-Functional Delivery: All-in-One Solution. Capable of effective delivery for plasmid DNA, siRNA, or simultaneous co-transfection of both nucleic acids.
- Broad Compatibility: Primary & Immortalised Cells. Specifically optimised to provide exceptional efficiency in many cell types, including challenging primary cells.
- Advanced Technology: Non-Liposomal. Utilises a proprietary polymeric system that avoids the toxicity often associated with liposomal reagents.
- Streamlined Workflow: No Media Changes. Fully compatible with serum-containing media; complexes are added directly to cells without subsequent wash steps.
- Robust Performance: High Efficiency. Delivers consistent, high-level gene expression or knockdown, supported by simple optimisation protocols.

Optimisation Guidelines
To ensure maximum efficiency, the TransIT-X2 system allows for flexible optimisation depending on the payload. For plasmid DNA transfection, the recommended starting condition is 3 µl of reagent per 1 µg of DNA, with an optimisation range of 2–6 µl. For siRNA delivery, a final concentration of 25 nM is recommended as a starting point, with a typical range of 10–50 nM. When performing co-transfections, the amount of reagent is dictated by the DNA quantity, ensuring a balanced delivery of both nucleic acids.
